In a person with MSA, an abnormal build-up of the protein\nα-synuclein is thought to be responsible for damaging the areas of the brain\nthat control balance, movement, and the body's normal functions.(1) MSA is\nseriously debilitating and places a high disease burden on patients. There is\ncurrently no cure for MSA and no available treatment to slow its clinical\nprogression.(2)\n\nSymptoms of MSA usually start between 55 and 60 years of age, and the typical\ntime to death is 8.6 years after symptom onset.(2 )Although there are many\ndifferent possible symptoms of MSA, not everyone who is affected will\nexperience all of them. The symptoms of MSA are wide-ranging and include\nmuscle control problems, similar to those of Parkinson's disease.(2) Many\ndifferent functions of the body can be affected, and symptoms including\nurinary incontinence, frequent falling, and unintelligible speech occur within\n3 years of disease onset. MSA is accompanied by reduced capacity to live\nindependently, and death is often due to respiratory problems.\n\nAbout amlenetug\nAmlenetug is a human monoclonal antibody (mAb) that recognizes and binds to\nall major forms of extracellular α-synuclein and thereby intended to prevent\nuptake and inhibit seeding of aggregation. Amlenetug is being developed by\nLundbeck under a joint research and licensing agreement between Lundbeck and\nGenmab A/S.\n\nAbout the MASCOT trial\nMASCOT (NCT06706622) is a Phase III interventional, randomized, double-blind,\nparallel-group, placebo-controlled, optional open-label extension trial that\nwill be conducted in North America, Europe, Asia and Australia.(3)\n\nThe trial comprises two parts: A double-blind period where participants are\nrandomized to receive either high or low doses of amlenetug, or placebo for 72\nweeks, followed by an open-label extension period where all participants\nenrolled in the trial are offered treatment with amlenetug. The aim of the\ntrial is to evaluate the efficacy, safety, and tolerability of amlenetug in\npatients with MSA. Amlenetug is delivered as an intravenous infusion every\nfour weeks.\n\nAbout Parkinson's disease\nParkinson's disease is a progressive and disabling neurological disease\ncharacterized by motor symptoms including slowness of movement, tremor and\nrigidity, as well as a range of non-motor symptoms.(4,5)\n\nThere are currently no established treatments that modify the progression of\nPD, and levodopa remains a cornerstone of symptomatic treatment.(6) With\nlong-term levodopa treatment, many people develop motor complications,\nincluding periods when symptoms return or worsen (\"OFF-time\") and dyskinesia\n(involuntary movements), which can significantly affect daily functioning and\nquality of life.(4-7)\n\nPD is the second most common neurodegenerative disease after Alzheimer's\ndisease and the most common movement disorder.(7-8 )In 2021, an estimated\n11.77 million people worldwide were living with PD, and its prevalence is\nprojected to more than double by 2050.(9) As PD progresses, functional\nindependence can decline and reliance on caregivers and healthcare systems can\nincrease, contributing to a substantial personal, societal and economic\nburden.\n\nAbout Lu AF28996\nLu AF28996 is an investigational, novel and oral prodrug of a\nD(1)-like/D(2)-like dopamine receptor agonist, with a pharmacologic profile\nthat may enhance dopaminergic striatal signaling in a prolonged manner. It is\nbeing investigated for its potential to improve motor fluctuations and\nlevodopa-induced dyskinesia in people with Parkinson's disease. Concerted\nactivation of D1- and D2-like dopamine receptors may play an important role in\nmotor control.\n\nLu AF28996 is currently in Phase II clinical development. The DARE2 trial\n(NCT07514858) is evaluating its efficacy, safety and tolerability in adults\nwith Parkinson's disease experiencing motor fluctuations despite optimized\nnon-invasive symptomatic treatment.(10)\n\nContacts\n Anders Crillsen Jens Høyer\n Senior Director, External & Internal Relations Vice President, Head of Investor Relations\n AECE@lundbeck.com (mailto:AECE@lundbeck.com) JSHR@lundbeck.com (mailto:JSHR@lundbeck.com)\n +45 27 79 12 86 +45 30 83 45 01\n\nAbout H.
