The findings were presented today in a\nLate-Breaking Oral Session at the International Gynecologic Cancer Society\n(IGCS) Congress 2026, held in Montreal, Canada.\n\nThe study also demonstrated a median progression-free survival (mPFS) of 9.5\nmonths (95% CI: 7.6–11.3). Antitumor activity was observed regardless of\nFRα expression levels, including in patients with low FRα expression and\nnon-expressors, and regardless of prior treatment with mirvetuximab.\n\n“Platinum-resistant ovarian cancer is a difficult-to-treat disease. As\npatients relapse and progress through successive lines of therapy, achieving\ndurable clinical benefit becomes increasingly challenging, yet remains\ncritically important,” said Elizabeth K. Lee, M.D., study investigator and a\nmedical oncologist in the gynecologic oncology program at Dana-Farber Cancer\nInstitute. “The antitumor activity and durability observed with Rina-S in\nthis heavily pretreated population are encouraging and suggest the potential\nto meaningfully impact outcomes for patients facing a particularly challenging\nstage of disease.”\n\nThe Part C cohort of the Phase 1/2 RAINFOL-01 study evaluated Rina-S 120\nmg/m² Q3W as monotherapy in patients with platinum-resistant high-grade\nserous ovarian, primary peritoneal, or fallopian tube cancer. Eligible\npatients received one to three prior lines of therapy; patients who received\nmirvetuximab as their last prior therapy could have received up to four prior\nlines. More than half of patients (53%) had received three or four prior lines\nof therapy. All patients had received prior bevacizumab and taxane therapy;\n49.5% had received a prior PARP inhibitor, and 33% had received prior\nmirvetuximab soravtansine. Median follow-up exceeded one year.\n\nThe most common treatment-emergent adverse events (TEAEs) included fatigue\n(57.8%) and low-grade (Grade 1-2) gastrointestinal events, such as nausea\n(67.9%), vomiting (36.7%), constipation (26.6%), decreased appetite (23.9%),\nand abdominal pain (18.3%). The most frequently reported hematologic TEAEs\nincluded anemia (57.8%), neutropenia (57.8%), decreased platelet count\n(34.9%), and thrombocytopenia (34.9%). Serious adverse events (SAEs) were\nreported in approximately one-third of participants. Treatment discontinuation\ndue to TEAEs occurred in 5.5% of participants. No safety signals for ocular\ntoxicity, peripheral neuropathy, interstitial lung disease (ILD), or\nstomatitis were observed.\n\n“The late-breaking results presented today add an important layer of\nevidence to the growing clinical experience with Rina-S in patients with\nplatinum-resistant ovarian cancer,” said Tahamtan Ahmadi, M.D., Ph.D.,\nExecutive Vice President and Chief Medical Officer, Head of Experimental\nMedicines, Genmab. “These findings, including the observed antitumor\nactivity, duration of response, and the first progression-free survival data\nreported for the program, as well as the manageable tolerability, reinforce\nthe potential of Rina-S. As our Phase 3 program continues to advance, the\nresults further inform our evaluation of Rina-S as a potential treatment\noption for patients with gynecologic cancers.”\n\nRina-S is being evaluated in a broad clinical development program spanning\nmultiple gynecologic cancers and other solid tumors. The program includes four\nPhase 3 trials evaluating Rina-S in PROC (RAINFOL-02; NCT06619236\n(https://cts.businesswire.com/ct/CT?id=smartlink&url=https%3A%2F%2Fclinicaltrials.gov%2Fstudy%2FNCT06619236&esheet=54614423&newsitemid=20261001541371&lan=en-US&anchor=NCT06619236&index=2&md5=437ef586368cd705d7640f1d0448ca72)\n), recurrent or progressive endometrial cancer (RAINFOL-03; NCT07166094\n(https://cts.businesswire.com/ct/CT?id=smartlink&url=https%3A%2F%2Fclinicaltrials.gov%2Fstudy%2FNCT07166094&esheet=54614423&newsitemid=20261001541371&lan=en-US&anchor=NCT07166094&index=3&md5=825f3b9b35eb864800f79abcd98dd635)\n), platinum-sensitive ovarian cancer (PSOC) maintenance therapy (RAINFOL-04;\nNCT07225270\n(https://cts.businesswire.com/ct/CT?id=smartlink&url=https%3A%2F%2Fclinicaltrials.gov%2Fstudy%2FNCT07225270&esheet=54614423&newsitemid=20261001541371&lan=en-US&anchor=NCT07225270&index=4&md5=3e9b8e1e20720f6996880ce069ead1bd)\n), and second-line PSOC (RAINFOL-07; NCT07564141\n(https://cts.businesswire.com/ct/CT?id=smartlink&url=https%3A%2F%2Fclinicaltrials.gov%2Fstudy%2FNCT07564141&esheet=54614423&newsitemid=20261001541371&lan=en-US&anchor=NCT07564141&index=5&md5=933ba283d3132f7f19ef92e42525a99a)\n). The study consists\nof multiple parts, including the PROC monotherapy cohort.\n\nAbout Ovarian Cancer\n\nOvarian cancer (OC) is a major global health issue, with more than 320,000 new\ncases diagnosed annually worldwide.(i) It ranks as the eighth most common\ncancer and the eighth leading cause of cancer-related deaths among women\nglobally.(ii) The disease is often diagnosed at an advanced stage because its\nsymptoms, including abdominal bloating, pelvic pain, and difficulty eating,\ncan be subtle and nonspecific.(iii) Approximately 70-90% of women with\nadvanced-stage OC worldwide experience a recurrence after initial\ntreatment.(iv )OC has a low five-year survival rate, which varies\nsignificantly by region, but generally hovers around 30-50%.(v,vi)\n\nAbout Rinatabart Sesutecan (Rina-S; GEN1184)\n\nRina-S (GEN1184) is an investigational ADC. It is composed of a novel human\nmonoclonal antibody directed at FRα, a hydrophilic protease-cleavable linker,\nand exatecan, a TOPO1 inhibitor payload. The clinical trial program for Rina-S\ncontinues to expand, including ovarian, endometrial, and other cancers with\nunmet need.\n\nThe safety and efficacy of Rina-S have not been established.
